    Landscapes are socially produced and reproduced spaces. This is easily recognizable for large-scale urban groups with built environments that dominate living places. But it also pertains to all types of societies and cultures, even small-scale hunter–gatherers, once the ontological beliefs structuring landscape perception and use are acknowledged. The foragers of south–central and southern California and the Great Basin illustrate this fact. They maintained a widely shared ontological perspective supported by a fundamental cognitive postulate. This is that supernatural power, the principle causative agent in the universe, was differentially distributed among individuals and places. The distribution of power, revealed by certain geomorphological features and natural events, structured their perceptions of landscape. These perceptions were expressed in ritual and symbolism, including petroglyphs and pictographs as durable manifestations of ceremonies on the landscape. The ontological relationship between power and landscape explains a longstanding question in hunter–gatherer archaeology: Why were rock writing sites created at specific locations? It also explains another equally significant but rarely considered and related problem: Why do some localities have massive quantities of rock writings that dwarf most other sites? The landscape symbolism of and the placement of sites by Native Californian and Great Basin tribes is explained by reference to their shared ontological beliefs, illustrating how they structured their ritual practices and archaeological record.

    A sacred site that draws pilgrims from distant regions is a distinctive resource for studying religion. Research into a site’s relevance to pilgrims and how it came to be founded contributes to a better understanding of religious activity. To address these issues, a thorough historical analysis of a sacred site’s records is essential. Such an analysis endeavors to distinguish the historical facts of a sacred site from its narratives and further discusses the significance of each. With such intent, this study focuses on the Rujing Stupa, a sacred site of significant importance to transnational pilgrimages that has yet to receive sufficient scholarly attention. The stupa, which is located at the Jingci Monastery in Hangzhou, China, is believed to hold the relics of Tiantong Rujing 天童如浄 (1163–1228), a Song Dynasty monk. Although the modern stele inscription at this location indicates that the stupa was founded in the 13th century, shortly after the monk’s death, this paper examines the historical reinventions within the inscription and traces the influence of Japanese narratives on such a reinvention. This study demonstrates that the Rujing Stupa was established by, and for, the Japanese Sōtō Buddhists. The Chinese monk’s connection to the Sōtō pilgrims lies in Rujing’s role as the master who instructed Dōgen 道元 (1200–1253), the founder of the Sōtō tradition, making his stupa a sacred site for the Sōtō community. Concerns of commemoration and reifying doctrinal authenticity motivated two generations of Japanese pilgrims to construct the Rujing stupa in the late 19th and 20th centuries, respectively. On the other hand, Rujing’s significance and the presence of the Sōtō tradition were scarcely acknowledged in China until the early modern period. Only in the late 20th century did Chinese Buddhists begin to appreciate this stupa. Examining the site’s historical reinventions and identifying the factors that shape its narrative, this case study offers insights into the investigation of sacred sites and suggests a concern for narrative in the examination of a site’s history and significance.

    AbstractIntroductionPosterior Cortical Atrophy (PCA) is a syndrome characterized by a progressive decline in higher-order visuospatial processing, leading to symptoms such as space perception deficit, simultanagnosia, and object perception impairment. While PCA is primarily known for its impact on visuospatial abilities, recent studies have documented language abnormalities in PCA patients. This study aims to delineate the nature and origin of language impairments in PCA, hypothesizing that language deficits reflect the visuospatial processing impairments of the disease.MethodsWe compared the language samples of 25 patients with PCA with age-matched cognitively normal (CN) individuals across two distinct tasks: a visually-dependent picture description and a visually-independent job description task. We extracted word frequency, word utterance latency, and spatial relational words for this comparison. We then conducted an in-depth analysis of the language used in the picture description task to identify specific linguistic indicators that reflect the visuospatial processing deficits of PCA.ResultsPatients with PCA showed significant language deficits in the visually-dependent task, characterized by higher word frequency, prolonged utterance latency, and fewer spatial relational words, but not in the visually-independent task. An in-depth analysis of the picture description task further showed that PCA patients struggled to identify certain visual elements as well as the overall theme of the picture. A predictive model based on these language features distinguished PCA patients from CN individuals with high classification accuracy.DiscussionThe findings indicate that language is a sensitive behavioral construct to detect visuospatial processing abnormalities of PCA. These insights offer theoretical and clinical avenues for understanding and managing PCA, underscoring language as a crucial marker for the visuospatial deficits of this atypical variant of Alzheimer’s disease.

    Although John Chrysostom is critical of the theatre, delivering a homily was never a tiresome monologue of the preacher in Byzantium; it was a theatrical performance combining text-reading and multiple ceremonies, during which spaces, lights, and materials were manipulated to create marvellous spectacles and enslave the audience spiritually and emotionally. At times, orators described the physical features of the venues where they spoke, as did Leo VI the Wise for two newly founded churches and Constantine VII Porphyrogenitus for the second most important church of the Empire, the Holy Apostles. But in most cases, the performance aspect of their speeches could only be known indirectly from two ceremonial handbooks, Kletorologion and De Ceremoniis. It is also necessary to indicate that the spectacles in homilies were not always real and present; they sometimes came to exist in listeners’ minds through picturesque descriptions (ekphraseis) and fictional figures (ethopoiiai) composed by preachers.

What is unclear from the narrative is the symbolic significance of the event, the materials used, the technical skills required, and whether magic was at play. Firstly, when considering the magical effects of the Nehushtan, we must define which type of magic—apotropaic or sympathetic—was involved. Based upon existing scholarship on the topic, the general consensus is that the Nehushtan represented sympathetic magic, underpinned in this instance by homeopathic/imitative magic. To highlight this point, this study will provide selected examples of both types of magic so that the Nehushtan’s association with sympathetic magic can be illustrated. Secondly, and most importantly, we must consider why the image of a snake was chosen if the very affliction (envenomation) suffered by the people was caused by the creature now being posited as a symbol of divine healing. Did the ancient perceptions of snakes and healing play a role in this decision? Why did the early Israelites not question the logic behind the use of a magical snake effigy when both magic and effigies were technically prohibited by biblical laws? To answer these questions, the study will consider the historical background (the Exodus from Egypt), the set (geographical location), and the setting (cultural contact and influence) in which the narrative of the Nehushtan took place.

    Information retrieval (IR) is essential in biomedical knowledge acquisition and clinical decision support. While recent progress has shown that language model encoders perform better semantic retrieval, training such models requires abundant query-article annotations that are difficult to obtain in biomedicine. As a result, most biomedical IR systems only conduct lexical matching. In response, we introduce MedCPT, a first-of-its-kind Contrastively Pre-trained Transformer model for zero-shot semantic IR in biomedicine. To train MedCPT, we collected an unprecedented scale of 255 million user click logs from PubMed. With such data, we use contrastive learning to train a pair of closely-integrated retriever and re-ranker. Experimental results show that MedCPT sets new state-of-the-art performance on six biomedical IR tasks, outperforming various baselines including much larger models such as GPT-3-sized cpt-text-XL. In addition, MedCPT also generates better biomedical article and sentence representations for semantic evaluations. As such, MedCPT can be readily applied to various real-world biomedical IR tasks.     Astrocytes not only support neuronal function with essential roles in synaptic neurotransmission, action potential propagation, metabolic support, or neuroplastic and developmental adaptations. They also respond to damage or dysfunction in surrounding neurons and oligodendrocytes by releasing neurotrophic factors and other molecules that increase the survival of the supported cells or contribute to mechanisms of structural and molecular restoration. The neuroprotective responsiveness of astrocytes is based on their ability to sense signals of degeneration, metabolic jeopardy and structural damage, and on their aptitude to locally deliver specific molecules to remedy threats to the molecular and structural features of their cellular partners. To the extent that neuronal and other glial cell disturbances are known to occur in affective disorders, astrocyte responsiveness to those disturbances may help to better understand the roles astrocytes play in affective disorders. The astrocytic sensing apparatus supporting those responses involves receptors for neurotransmitters, purines, cell adhesion molecules and growth factors. Astrocytes also share with the immune system the capacity of responding to cytokines released upon neuronal damage. In addition, in responses to specific signals astrocytes release unique factors such as clusterin or humanin that have been shown to exert potent neuroprotective effects. Astrocytes integrate the signals above to further deliver structural lipids, removing toxic metabolites, stabilizing the osmotic environment, normalizing neurotransmitters, providing anti-oxidant protection, facilitating synaptogenesis and acting as barriers to contain varied deleterious signals, some of which have been described in brain regions relevant to affective disorders and related animal models. Since various of the injurious signals that activate astrocytes have been implicated in different aspects of the etiopathology of affective disorders, particularly in relation to the diagnosis of depression, potentiating the corresponding astrocyte neuroprotective responses may provide additional opportunities to improve or complement available pharmacological and behavioral therapies for affective disorders.

    The history of adrenoleukodystrophy (ALD), adrenomyeloneuropathy (AMN) and other peroxisomal diseases is exemplary for the stunning progress of scientific medicine within the past 50 years. Like many breakthroughs in medicine, the detailed analysis of patients’ pathologically affected tissues was instrumental, resulting in step-wise systematic clarification of what had remained enigmatic until the 1970s. This flashback paper is a recollection of the first neuropathological description of a slowly evolving clinical phenotype, spastic paraparesis with adrenal insufficiency, in a young adult by Budka et al. 1976 [3], using virtual microscopy of the original histologic slides. The clinico-pathological presentation derives from the classical cerebral ALD phenotype in boys, where electron microscopy demonstrated the underlying pathological hallmark of characteristic lipid inclusions shared by both phenotypes. Our report allowed the delineation of a new disease type almost simultaneously described in more cases as AMN by Griffin et al. 1977 [4] and Schaumburg et al. 1977 [11]. Moreover, our report indicated clinical heterogeneity in the ALD disease group that, as shown later, extends further to females, to Addison-only, and even to asymptomatic subjects. The gene underlying ALD was discovered in 1993 as a defect in the ABCD1 gene. Yet, it has hitherto remained unclear how the gene defect causes the strikingly broad and unpredictable phenotypic spectrum of ALD/AMN. Free Neuropathology, Bd. 4 (2023)

    Age is the greatest risk factor for the development of type 2 diabetes mellitus (T2DM). Age-related decline in organ function is attributed to the accumulation of stochastic damage, including damage to the nuclear genome. Islets of T2DM patients display increased levels of DNA damage. However, whether this is a cause or consequence of the disease has not been elucidated. Here, we asked if spontaneous, endogenous DNA damage in β-cells can drive β-cell dysfunction and diabetes, via deletion of Ercc1, a key DNA repair gene, in β-cells. Mice harboring Ercc1-deficient β-cells developed adult-onset diabetes as demonstrated by increased random and fasted blood glucose levels, impaired glucose tolerance, and reduced insulin secretion. The inability to repair endogenous DNA damage led to an increase in oxidative DNA damage and apoptosis in β-cells and a significant loss of β-cell mass. Using electron microscopy, we identified β-cells in clear distress that showed an increased cell size, enlarged nuclear size, reduced number of mature insulin granules, and decreased number of mitochondria. Some β-cells were more affected than others consistent with the stochastic nature of spontaneous DNA damage. Ercc1-deficiency in β-cells also resulted in loss of β-cell function as glucose-stimulated insulin secretion and mitochondrial function were impaired in islets isolated from mice harboring Ercc1-deficient β-cells. These data reveal that unrepaired endogenous DNA damage is sufficient to drive β-cell dysfunction and provide a mechanism by which age increases the risk of T2DM.

    Landscapes are socially produced and reproduced spaces. This is easily recognizable for large-scale urban groups with built environments that dominate living places. But it also pertains to all types of societies and cultures, even small-scale hunter–gatherers, once the ontological beliefs structuring landscape perception and use are acknowledged. The foragers of south–central and southern California and the Great Basin illustrate this fact. They maintained a widely shared ontological perspective supported by a fundamental cognitive postulate. This is that supernatural power, the principle causative agent in the universe, was differentially distributed among individuals and places. The distribution of power, revealed by certain geomorphological features and natural events, structured their perceptions of landscape. These perceptions were expressed in ritual and symbolism, including petroglyphs and pictographs as durable manifestations of ceremonies on the landscape. The ontological relationship between power and landscape explains a longstanding question in hunter–gatherer archaeology: Why were rock writing sites created at specific locations? It also explains another equally significant but rarely considered and related problem: Why do some localities have massive quantities of rock writings that dwarf most other sites? The landscape symbolism of and the placement of sites by Native Californian and Great Basin tribes is explained by reference to their shared ontological beliefs, illustrating how they structured their ritual practices and archaeological record.

    A sacred site that draws pilgrims from distant regions is a distinctive resource for studying religion. Research into a site’s relevance to pilgrims and how it came to be founded contributes to a better understanding of religious activity. To address these issues, a thorough historical analysis of a sacred site’s records is essential. Such an analysis endeavors to distinguish the historical facts of a sacred site from its narratives and further discusses the significance of each. With such intent, this study focuses on the Rujing Stupa, a sacred site of significant importance to transnational pilgrimages that has yet to receive sufficient scholarly attention. The stupa, which is located at the Jingci Monastery in Hangzhou, China, is believed to hold the relics of Tiantong Rujing 天童如浄 (1163–1228), a Song Dynasty monk. Although the modern stele inscription at this location indicates that the stupa was founded in the 13th century, shortly after the monk’s death, this paper examines the historical reinventions within the inscription and traces the influence of Japanese narratives on such a reinvention. This study demonstrates that the Rujing Stupa was established by, and for, the Japanese Sōtō Buddhists. The Chinese monk’s connection to the Sōtō pilgrims lies in Rujing’s role as the master who instructed Dōgen 道元 (1200–1253), the founder of the Sōtō tradition, making his stupa a sacred site for the Sōtō community. Concerns of commemoration and reifying doctrinal authenticity motivated two generations of Japanese pilgrims to construct the Rujing stupa in the late 19th and 20th centuries, respectively. On the other hand, Rujing’s significance and the presence of the Sōtō tradition were scarcely acknowledged in China until the early modern period. Only in the late 20th century did Chinese Buddhists begin to appreciate this stupa. Examining the site’s historical reinventions and identifying the factors that shape its narrative, this case study offers insights into the investigation of sacred sites and suggests a concern for narrative in the examination of a site’s history and significance.

    AbstractIntroductionPosterior Cortical Atrophy (PCA) is a syndrome characterized by a progressive decline in higher-order visuospatial processing, leading to symptoms such as space perception deficit, simultanagnosia, and object perception impairment. While PCA is primarily known for its impact on visuospatial abilities, recent studies have documented language abnormalities in PCA patients. This study aims to delineate the nature and origin of language impairments in PCA, hypothesizing that language deficits reflect the visuospatial processing impairments of the disease.MethodsWe compared the language samples of 25 patients with PCA with age-matched cognitively normal (CN) individuals across two distinct tasks: a visually-dependent picture description and a visually-independent job description task. We extracted word frequency, word utterance latency, and spatial relational words for this comparison. We then conducted an in-depth analysis of the language used in the picture description task to identify specific linguistic indicators that reflect the visuospatial processing deficits of PCA.ResultsPatients with PCA showed significant language deficits in the visually-dependent task, characterized by higher word frequency, prolonged utterance latency, and fewer spatial relational words, but not in the visually-independent task. An in-depth analysis of the picture description task further showed that PCA patients struggled to identify certain visual elements as well as the overall theme of the picture. A predictive model based on these language features distinguished PCA patients from CN individuals with high classification accuracy.DiscussionThe findings indicate that language is a sensitive behavioral construct to detect visuospatial processing abnormalities of PCA. These insights offer theoretical and clinical avenues for understanding and managing PCA, underscoring language as a crucial marker for the visuospatial deficits of this atypical variant of Alzheimer’s disease.

    Although John Chrysostom is critical of the theatre, delivering a homily was never a tiresome monologue of the preacher in Byzantium; it was a theatrical performance combining text-reading and multiple ceremonies, during which spaces, lights, and materials were manipulated to create marvellous spectacles and enslave the audience spiritually and emotionally. At times, orators described the physical features of the venues where they spoke, as did Leo VI the Wise for two newly founded churches and Constantine VII Porphyrogenitus for the second most important church of the Empire, the Holy Apostles. But in most cases, the performance aspect of their speeches could only be known indirectly from two ceremonial handbooks, Kletorologion and De Ceremoniis. It is also necessary to indicate that the spectacles in homilies were not always real and present; they sometimes came to exist in listeners’ minds through picturesque descriptions (ekphraseis) and fictional figures (ethopoiiai) composed by preachers.

    According to Numbers 21:4–9, the Nehushtan was a copper/bronze snake effigy that functioned as a ‘magical’ healing tool to cure the early Israelites from venomous snakebites they incurred during their desert wanderings. What is unclear from the narrative is the symbolic significance of the event, the materials used, the technical skills required, and whether magic was at play. Firstly, when considering the magical effects of the Nehushtan, we must define which type of magic—apotropaic or sympathetic—was involved. Based upon existing scholarship on the topic, the general consensus is that the Nehushtan represented sympathetic magic, underpinned in this instance by homeopathic/imitative magic. To highlight this point, this study will provide selected examples of both types of magic so that the Nehushtan’s association with sympathetic magic can be illustrated. Secondly, and most importantly, we must consider why the image of a snake was chosen if the very affliction (envenomation) suffered by the people was caused by the creature now being posited as a symbol of divine healing. Did the ancient perceptions of snakes and healing play a role in this decision? Why did the early Israelites not question the logic behind the use of a magical snake effigy when both magic and effigies were technically prohibited by biblical laws? To answer these questions, the study will consider the historical background (the Exodus from Egypt), the set (geographical location), and the setting (cultural contact and influence) in which the narrative of the Nehushtan took place.

    Information retrieval (IR) is essential in biomedical knowledge acquisition and clinical decision support. While recent progress has shown that language model encoders perform better semantic retrieval, training such models requires abundant query-article annotations that are difficult to obtain in biomedicine. As a result, most biomedical IR systems only conduct lexical matching. In response, we introduce MedCPT, a first-of-its-kind Contrastively Pre-trained Transformer model for zero-shot semantic IR in biomedicine. To train MedCPT, we collected an unprecedented scale of 255 million user click logs from PubMed. With such data, we use contrastive learning to train a pair of closely-integrated retriever and re-ranker. Experimental results show that MedCPT sets new state-of-the-art performance on six biomedical IR tasks, outperforming various baselines including much larger models such as GPT-3-sized cpt-text-XL. In addition, MedCPT also generates better biomedical article and sentence representations for semantic evaluations. As such, MedCPT can be readily applied to various real-world biomedical IR tasks.     Astrocytes not only support neuronal function with essential roles in synaptic neurotransmission, action potential propagation, metabolic support, or neuroplastic and developmental adaptations. They also respond to damage or dysfunction in surrounding neurons and oligodendrocytes by releasing neurotrophic factors and other molecules that increase the survival of the supported cells or contribute to mechanisms of structural and molecular restoration. The neuroprotective responsiveness of astrocytes is based on their ability to sense signals of degeneration, metabolic jeopardy and structural damage, and on their aptitude to locally deliver specific molecules to remedy threats to the molecular and structural features of their cellular partners. To the extent that neuronal and other glial cell disturbances are known to occur in affective disorders, astrocyte responsiveness to those disturbances may help to better understand the roles astrocytes play in affective disorders. The astrocytic sensing apparatus supporting those responses involves receptors for neurotransmitters, purines, cell adhesion molecules and growth factors. Astrocytes also share with the immune system the capacity of responding to cytokines released upon neuronal damage. In addition, in responses to specific signals astrocytes release unique factors such as clusterin or humanin that have been shown to exert potent neuroprotective effects. Astrocytes integrate the signals above to further deliver structural lipids, removing toxic metabolites, stabilizing the osmotic environment, normalizing neurotransmitters, providing anti-oxidant protection, facilitating synaptogenesis and acting as barriers to contain varied deleterious signals, some of which have been described in brain regions relevant to affective disorders and related animal models. Since various of the injurious signals that activate astrocytes have been implicated in different aspects of the etiopathology of affective disorders, particularly in relation to the diagnosis of depression, potentiating the corresponding astrocyte neuroprotective responses may provide additional opportunities to improve or complement available pharmacological and behavioral therapies for affective disorders.

    The history of adrenoleukodystrophy (ALD), adrenomyeloneuropathy (AMN) and other peroxisomal diseases is exemplary for the stunning progress of scientific medicine within the past 50 years. Like many breakthroughs in medicine, the detailed analysis of patients’ pathologically affected tissues was instrumental, resulting in step-wise systematic clarification of what had remained enigmatic until the 1970s. This flashback paper is a recollection of the first neuropathological description of a slowly evolving clinical phenotype, spastic paraparesis with adrenal insufficiency, in a young adult by Budka et al. 1976 [3], using virtual microscopy of the original histologic slides. The clinico-pathological presentation derives from the classical cerebral ALD phenotype in boys, where electron microscopy demonstrated the underlying pathological hallmark of characteristic lipid inclusions shared by both phenotypes. Our report allowed the delineation of a new disease type almost simultaneously described in more cases as AMN by Griffin et al. 1977 [4] and Schaumburg et al. 1977 [11]. Moreover, our report indicated clinical heterogeneity in the ALD disease group that, as shown later, extends further to females, to Addison-only, and even to asymptomatic subjects. The gene underlying ALD was discovered in 1993 as a defect in the ABCD1 gene. Yet, it has hitherto remained unclear how the gene defect causes the strikingly broad and unpredictable phenotypic spectrum of ALD/AMN. Free Neuropathology, Bd. 4 (2023)

    Age is the greatest risk factor for the development of type 2 diabetes mellitus (T2DM). Age-related decline in organ function is attributed to the accumulation of stochastic damage, including damage to the nuclear genome. Islets of T2DM patients display increased levels of DNA damage. However, whether this is a cause or consequence of the disease has not been elucidated. Here, we asked if spontaneous, endogenous DNA damage in β-cells can drive β-cell dysfunction and diabetes, via deletion of Ercc1, a key DNA repair gene, in β-cells. Mice harboring Ercc1-deficient β-cells developed adult-onset diabetes as demonstrated by increased random and fasted blood glucose levels, impaired glucose tolerance, and reduced insulin secretion. The inability to repair endogenous DNA damage led to an increase in oxidative DNA damage and apoptosis in β-cells and a significant loss of β-cell mass. Using electron microscopy, we identified β-cells in clear distress that showed an increased cell size, enlarged nuclear size, reduced number of mature insulin granules, and decreased number of mitochondria. Some β-cells were more affected than others consistent with the stochastic nature of spontaneous DNA damage. Ercc1-deficiency in β-cells also resulted in loss of β-cell function as glucose-stimulated insulin secretion and mitochondrial function were impaired in islets isolated from mice harboring Ercc1-deficient β-cells. These data reveal that unrepaired endogenous DNA damage is sufficient to drive β-cell dysfunction and provide a mechanism by which age increases the risk of T2DM.
